Gebaut wurde die Stadt von Seleukos, einem Feldherren Alexander des Großen, der der frisch gegründeten Stadt den Namen seiner Frau Apamea gab. In römischer Zeit wohnten fast eine halbe Million Menschen in der Stadt, die damals Pella genannt wurde. Im zweiten Jahrhundert nach Christus wurde die mit 1600 m Länge und 40 m Breite die längste römische Säulenstraße des römischen Imperiums gebaut, deren Teile bis heute zu sehen sind. Im vierten Jahrhundert wurde Apamea Bischoffsitz, im 6. Jh eroberten die Perser und bald darauf die Muslime die Stadt. Apamea verlor im Laufe der Jahrhunderte immer mehr an Bedeutung. Die Kreuzfahrer verhalfen der Stadt noch einmal zu ein bisschen Glanz, als sie sich im 12. Jh in der Zitadelle des Seleukos niederließen, die hoch über dem Orontes auf einem Hügel trohnt, doch nach der Eroberung durch Nur ed-Din 1149 wurde die Stadt 1157 durch ein Erdbeben völlig zerstört.
